Clonal Expansion of Lymphocytes Bearing the ‘y#{246} T-Cell Receptor in a Patient With Large Granular Lymphocyte Disorder

چکیده
Repeated analysis of peripheral blood lymphocytes (PBLs) from a patient with large granular lymphocytosis. neutropenia, and rheumatoid arthritis revealed that -45% of PBLs displayed the following phenotype: CD3 , CD4, CDS. CD1 6 . HNK-1 -, WT31 -. This population was purified for further analysis by depletion with anti-CD4 and anti-CD8 monoclonal antibodies (MoAbs). Southern blot analysis showed preferential rearrangements of the Vy9 genes. Northern blot demonstrated the presence of Vy9 mRNA transcripts. With MoAbs directed against either the V’y9 peptide (Ti’yA) or the #{246} chain of the ‘y#{246} T-cell receptor

Detection of monoclonal T populations in patients with KIR-restricted chronic lymphoproliferative disorder of NK cells.
The etiology of chronic large granular lymphocyte proliferations is largely unknown. Although these disorders are characterized by the expansion of different cell types (T and natural killer) with specific genetic features and abnormalities, several lines of evidence suggest a common pathogenetic mechanism. According to this interpretation, we speculated that in patients with natural killer-typ...
متن کاملT cell receptor beta chain gene rearrangements in lymphoproliferative disorders of large granular lymphocytes/natural killer cells
Twelve cases of T gamma LPD (lymphoproliferative disorders of Fc gamma receptor-bearing T cells) involving an expansion of large granular lymphocyte/natural killer (LGL/NK) cells were investigated for the expression of LGL/NK-associated markers and for T beta gene rearrangement. All the cases selected were classified as T gamma LPD on the basis of morphology, function, and phenotype of the circ...

A 43-year-old male with a phenotypically homogeneous, expanded subset of T cells presented in 1981 with anemia and neutropenia. The surface antigen phenotype of 99% of the peripheral blood lymphocytes was T3+, T8+, T4-, and they were morphologically large granular lymphocytes (LGL). The same cells comprised 37% of the marrow nucleated cells. Eight months after he presented, the peripheral blood...

A case with the triad of pure red cell aplasia (PRCA), myasthenia gravis, and malignant thymoma is reported. There was a clonal proliferation of T cells within the thymoma, as demonstrated by a T cell antigen receptor (TCR) delta chain gene rearrangement. However, despite a large granular lymphocytosis, clonality could not be shown in the peripheral blood either before or after thymectomy. Ther...
متن کاملClonal drift demonstrates unexpected dynamics of the T-cell repertoire in T-large granular lymphocyte leukemia.
T-cell large granular lymphocyte leukemia (T-LGLL) is characterized by chronic lymphoproliferation of cytotoxic T lymphocytes (CTLs) and is associated with lineage-restricted cytopenias. Introduction of T-cell receptor (TCR) variable β-chain (Vβ) monoclonal antibodies has facilitated identification and enumeration of clonal CTLs by flow cytometry. A highly skewed TCR Vβ repertoire identified by...
